The short answer

What is NAD+?

NAD+ is nicotinamide adenine dinucleotide, a coenzyme present in every cell and one of the oldest molecules in biochemistry. It has two jobs in the published account. The first is redox: it shuttles electrons between the reactions that release energy from fuel, cycling between an oxidized and a reduced form without being used up. The second is as a consumed substrate, because sirtuins and PARP enzymes cleave NAD+ as part of their normal signaling, which ties the coenzyme's supply to those pathways (Covarrubias et al., 2021). Reviews of NAD+ metabolism describe tissue levels falling with age, which is the observation that turned a textbook cofactor into a research target (Covarrubias et al., 2021; Rajman et al., 2018). Peptara supplies research-grade NAD+ as a lyophilized 1000mg vial for laboratory research; this page describes what published reviews report, not an effect in any person, and a qualified clinician is the only place a personal question belongs.

The Science

A Cofactor That Is Also a Currency

Most cofactors are recycled. NAD+ is recycled for its redox work and spent for its signaling work, and that second role is what makes its supply a variable rather than a constant. The published literature is built around that distinction.

Redox Carrier

Cycles between oxidized and reduced forms, carrying electrons through energy metabolism.

Signaling Substrate

Cleaved rather than returned by sirtuins and PARP enzymes, so those pathways draw on the pool.

Falls With Age

Reviews describe tissue levels declining over time across model systems.

Applications

Common Research Areas

Energy Metabolism

The redox role in the reactions that release energy from fuel is the best established part of the record.

Sirtuin Biology

Sirtuins consume NAD+, so the coenzyme appears throughout that literature as a dependency.

DNA Damage Response

PARP enzymes also cleave NAD+, which is why DNA repair work touches the same pool.

Age-Related Decline

The reported fall in tissue levels with age is the research area, not a demonstrated intervention.

Naming

NAD+, NMN and NR: what the names mean

These three are routinely treated as interchangeable and they are not. NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside) are intermediates in the salvage pathway that builds NAD+. They are precursors. NAD+ is the coenzyme itself, the molecule the redox and signaling reactions actually use. Yoshino and colleagues set the two apart explicitly, reviewing the biology of the intermediates as their own subject with their own evidence base (Yoshino et al., 2018).

The distinction matters when reading a study. A trial of NR is a trial of a precursor and its absorption, conversion and downstream effect; it is not a direct measurement of NAD+ itself. What ships here is NAD+ as a lyophilized 1000mg vial, so papers about NMN or NR describe a related molecule rather than this one, and this page does not borrow their results.

Evidence

How strong is the NAD+ evidence?

The mechanistic account is well established. NAD+ metabolism, the enzymes that consume it, and the reported decline in tissue levels with age are all covered in detail in the review literature (Covarrubias et al., 2021). That is the settled part.

The unsettled part is whether raising NAD+ changes an outcome. Rajman and colleagues reviewed the in vivo evidence for NAD-boosting molecules and were explicit about the shape of it: substantial animal work, and a human record that is smaller and earlier than the attention the field receives (Rajman et al., 2018). This page reports that gap rather than writing over it. No result here is an outcome claim, and nothing on this page is medical advice.

The certificate for this batch is pending. Fourteen of the nineteen catalog SKUs carry a published third-party panel today, listed on the certificates page, and the remaining five say pending rather than showing a figure no lab has measured.

What the reviews describe, step by step

The coenzyme

NAD+ is nicotinamide adenine dinucleotide, a redox coenzyme that carries electrons through the reactions of energy metabolism in every cell (Covarrubias et al., 2021).

The second job

It is also consumed as a substrate rather than only recycled: sirtuins and PARP enzymes cleave NAD+ as part of their normal signaling, which links its supply to those pathways (Covarrubias et al., 2021).

The decline

Published reviews describe tissue NAD+ falling with age across model systems, which is why the coenzyme became a longevity research target at all (Covarrubias et al., 2021; Rajman et al., 2018).

The open question

Rajman and colleagues reviewed the in vivo evidence for NAD-boosting molecules and set out what animal work has shown against what human work has not yet settled (Rajman et al., 2018).
