How P21 works in research models
BDNF in animal studies
Aged-rat work measured higher BDNF, a growth signal for nerve cells. The same result did not hold in the 2024 CDKL5 mouse model.
Tau changes in mice
An Alzheimer mouse study measured lower changes in tau, a protein linked to the model, and lower soluble amyloid beta.
New nerve cells
The first mouse paper measured the birth and maturing of new nerve cells in the hippocampus, a brain area involved in memory.

The 2024 CDKL5 study found that P021 restored nerve-cell growth and survival in a dish. In mice, it did not raise BDNF or correct brain structure changes, and behavior changes were limited.
The founding 2010 and 2014 papers came from the New York State Institute for Basic Research in Developmental Disabilities in Staten Island. The 2017 Down syndrome-model paper states that its senior author holds U.S. patents on CNTF small-molecule peptide mimetics. The shared institute and declared patent interest belong alongside the results when weighing this evidence.
